The Victoria swim star finished 11th in qualifying at the Olympic Aquatics Stadium in Rio de Janeiro on Saturday in a time of three minutes, 45.83 seconds, more than two seconds behind his Canadian record of 3:43.46.
"It's frustrating, but I know I'm fortunate enough to have two chances at the Olympics and not a lot of people get that second chance," said Cochrane. "I'm an old-enough athlete to know where to see what I need to work on and I have six days to do that."
Saturday's eight-man final is scheduled for 9:30 p.m. ET.
At the 2012 London Olympics, a tactical error during qualifying cost Cochrane a spot in the final. He missed by one-hundredth of a second when gold medallist Park Tae-hwan had his disqualification overturned.
A two-time Olympic medallist, Cochrane will also race the 1,500 – his stronger event – in Brazil on Friday. Cochrane won bronze in the 1,500 at the 2008 Beijing Games and silver in the same event in London.
He is the most-decorated swimmer in Canadian history with eight world championship medals, including bronze in both disciplines at last year's FINA world championships in Kazan, Russia.
Women's 400 IM
A bronze-medal winner at last year's world championships, Canada's Emily Overholt will have a chance at Olympic gold.
The Vancouver native secured the eighth and final spot for Saturday's final at 9:49 p.m. with a 4:36.54 clocking to finish third in her heat. Overholt has the Canadian women's record of 4:32.52.
Teammate Sydney Pickrem wasn't as lucky. While her 4:38.06 performance was narrowly off her Olympic qualifying time of 4:38.03, it wasn't good enough to advance. She placed 12th.
Women's 4x100 free relay
The Canadian foursome of Sandrine Mainville, Chantal van Landeghem, Michelle Williams and Taylor Ruck finished third in their heat in 3:33.84 to qualify third for the eight-team final on Saturday at 10:24 p.m.
Australia was first, setting an Olympic record of 3:32.39.
Women's 100 fly
Canada's Penny Oleksiak, 16, was outside the top three after 50 metres but demonstrated her explosive closing speed, finishing second in her heat and third overall in a time of 56.73 seconds.
The six-foot-one Toronto swimmer, competing at her first Olympics, advances to Saturday's semifinals at 9:14 p.m. ET. The final is Sunday at 9:03 p.m.
Noemi Thomas of Richmond, B.C., led halfway through her heat but faded over the final 50 metres, touching the wall in 58.27 and failed to advance to the semifinal field of 15. She finished in a tie for 18th.
Men's 100 breaststroke
Calgary's Jason Block placed third in his heat in 1:00.71 but couldn't crack the 15-man semifinal, set for 10:08 p.m. Saturday. He finished atop the podium at the Canadian Olympic trials in April with a time of 1:00.59.
Great Britain's Adam Peaty qualified first, stopping the clock in a world-record 57.55 seconds.
